چکیده

What is it? The Mediator complex is an enormous, multi-subunit protein complex that regulates gene expression in eukaryotic cells. It acts as a molecular circuit board that integrates physiological and developmental signals and transmits them to RNA polymerase II (Pol II), the enzyme charged with transcription of all protein-encoding genes. Although initially discovered as a cofactor required for activatordependent transcription, Mediator also stimulates activator-independent (basal) transcription. Furthermore, Mediator can act as a co-repressor that facilitates transcriptional silencing. Mediator activity is not limited to its effects on transcription initiation, but it can also affect elongation and perhaps all stages of the transcription cycle.

Mediator Complex Subunit 12 Gene Polymorphisms in Uterine Fibroids and Breast Fibroadenomas in Senegalese Women

Mediator complex subunit 12 (MED12) is a part of the mediator complex, which is believed to regulate transcription. MED12 is mutated at high frequency and with different mutation frequencies in uterine fibroids and breast fibroadenomas of different populations. This study aimed to analyze MED12 mutations in Senegalese population. MED12 was sequenced in the tumoral tissues and blood samples of S...

The tail-module of yeast Mediator complex is required for telomere heterochromatin maintenance

Eukaryotic chromosome ends have a DNA-protein complex structure termed telomere. Integrity of telomeres is essential for cell proliferation. Genome-wide screenings for telomere length maintenance genes identified several components of the transcriptional regulator, the Mediator complex. Our work provides evidence that Mediator is involved in telomere length regulation and telomere heterochromat...

Mediator Undergoes a Compositional Change during Transcriptional Activation.

Mediator is a transcriptional co-activator recruited to enhancers by DNA-binding activators, and it also interacts with RNA polymerase (Pol) II as part of the preinitiation complex (PIC). We demonstrate that a single Mediator complex associates with the enhancer and core promoter in vivo, indicating that it can physically bridge these transcriptional elements. However, the Mediator kinase modul...

Transcriptional activation: mediator can act after preinitiation complex formation.

The Mediator complex connects transcriptional activators bound at enhancers with RNA polymerase (Pol) II. demonstrate that Mediator also has an important role in transcriptional activation after recruitment of the Pol II machinery to promoters.

Regulation of metabolism by the Mediator complex

The Mediator complex was originally discovered in yeast, but it is conserved in all eukaryotes. Its best-known function is to regulate RNA polymerase II-dependent gene transcription. Although the mechanisms by which the Mediator complex regulates transcription are often complicated by the context-dependent regulation, this transcription cofactor complex plays a pivotal role in numerous biologic...

The Mediator complex and transcription regulation

The Mediator complex is a multi-subunit assembly that appears to be required for regulating expression of most RNA polymerase II (pol II) transcripts, which include protein-coding and most non-coding RNA genes. Mediator and pol II function within the pre-initiation complex (PIC), which consists of Mediator, pol II, TFIIA, TFIIB, TFIID, TFIIE, TFIIF and TFIIH and is approximately 4.0 MDa in size...
